The problem is to make the riddle work you have to resort to bad grammar. If referring to the letters in the words you wouldn’t use the article ‘a’ before the noun. It’s just bad English but to use correct English ruins the riddle as it would go:
Two in corner
One in room
Zero in house but one in shelter.
